一般的sp法步骤啊，具体如下：
1、烤片，68℃，20分钟，
2、常规二甲苯脱蜡，梯度酒精脱水；二甲苯I20min?二甲苯II20min?100%酒精10min?100%酒精10min?95%酒精5min?80%酒精5min?70%酒精5min
3、阻断灭活内源性过氧化物酶：3%H2O237℃孵育10min，PBS冲洗3X5min；
4、抗原修复：置0.01M枸橼酸缓冲液（PH6.0）中用煮沸（95℃，15-20min），自然冷却20min以上，再用冷水冲洗缸子，加快冷却至室温，PBS冲洗3X5min。
5、正常羊血清工作液封闭，37℃10min，倾去勿洗。
6、滴加一抗4℃冰箱孵育过夜，PBS冲洗3X5min（用PBS缓冲液代替一抗作阴性对照）；
7、滴加生物素标记二抗，37℃孵育30min，PBS冲洗3X5min；
8、滴加辣根过氧化物酶标记的链霉素卵白素工作液，37℃孵育30min，PBS冲洗3X5min；
9、DAB/H2O2反应染色，自来水充分冲洗后，苏木素复染，常规脱水，透明，干燥，封片。

General sp method steps ah, as follows:

4. Antigen Recover: Set 0.01M citrate buffer (PH6.0) with boiling (95 ℃, 15-20min) for more than 20min, then rinse the cylinder with cold water, cool to room temperature and rinse with PBS for 3X5min.
5, normal sheep serum working fluid closed, 37 ℃ 10min, do not dump.
6, dropping the first antibody incubated overnight at 4 ℃ refrigerator, PBS wash 3X5min (PBS buffer instead of primary antibody as a negative control);
7, dropping biotin labeled secondary antibody, 37 ℃ incubated 30min, PBS wash 3X5min;
8, dropping horseradish peroxidase labeled streptomycin avidin working solution, incubated at 37 ℃ 30min, PBS washed 3X5min;
9, DAB / H2O2 reaction staining, tap water fully washed, hematoxylin counterstain, conventional dehydration, transparent, dry, sealing pieces.
