利用药剂迅速把细胞杀死，以保持材料本来的状态和结构。
常用的固定有F.A.A固定液、卡诺固定液等。将固定液放在具塞小瓶中，投入材料，盖好瓶盖。用F.A.A固定液固定，时间至少24小时。F.A.A固定液既是良好的固定剂，也是保存剂，因此材料可长期保存在固定液中备用。卡诺固定液穿透力强，固定较小材料一般1~6小时即可，zui多不超过24小时。因卡诺固定液不能做保存剂，所以固定后要用95%、85%酒精浸洗，每级约20min.，然后保存在70%的酒精中备用。
（三）脱水
脱水的目的是除去组织中的水分，便于透明、浸蜡、包埋等步骤的进行，因为这些程序中所用的药品都不能与水混合，所以必须脱水。常用的脱水剂为酒精。脱水过程应缓慢进行，不能操之过急或时间过长，以免组织变硬变脆或发生收缩现象。脱水采用等级脱水（系列脱水），即从较低浓度酒精开始，逐渐替换到高浓度酒精。一般为：50%→70%→80%→95%（此时可加少许番红，使组织着色便于包埋时定位）→*（两次），每级停留时间2~4小时或更长（*中时间宜短）。
用95%酒精配制各浓度酒精的方法：设原有酒精浓度为A，所需配制酒精浓度为B，则取Bml原有酒精，加上A-Bml蒸馏水，即可获得Aml的B%的究竟。例如：95%酒精配成70%的酒精，则取70ml的95%酒精，加上25ml蒸馏水，则配成了95ml的70%的酒精。
