4、免疫组化中抗原修复的*条件是什么？（尤其是微波修复）
关于抗原修复，我试过的修复条件有EDTA热修复，柠檬酸钠为缓冲液的微波修复以及高压锅修复。从我的实验结果来看，微波修复其实很不容易掉片子的，而EDTA热修复和高压锅修复是很容易掉片子的。因为掉片子主要是因为加热过程中产生的气泡所引起，而微波修复水加热到沸腾，沾在片子上的气泡就会少，不会因为小气泡上串引起脱片。做EDTA修复时，你可以将片子靠的尽量近些，或是在载玻片四周垫些棉花什么的，然后盖上盖玻片，这样修复的话就能够尽量减少载玻片上小气泡的形成。我们用的微波修复条件为：2.94 g柠檬酸三钠溶于1000 ml的水，调PH值至6.0，微波修复10分钟。你可以试试，时间可以根据需要自己调整。对于柠檬酸修复来说，只要高压修复时间控制在加阀冒气后90秒，冷水下终止开高压锅盖，高压修复和微波修复一样不掉片子，而且一般高压修复效果往往更好一点；关于EDTA热修复，一般说明书上都讲的是PH值等于9，实际上我试过，如果单传用EDTA，很容易掉片子，但是用Tris-EDTA,一般就不会掉片子了，Tris-base的浓度为0.05 mmol/L,高压和微波修复都可以，PH也等于9。
