9．封闭：在山羊血清封闭，非特异性染色仍然较强时，可延长封闭时间或用浓缩血清封闭
10．抗体稀释：应遵循“现用现配”的原则，对于PBS稀释的抗体一定要当天使用。
11．背景高：在抗体浓度、反应时间、反应温度等合适的条件下，如果背景依旧高，可采用含1‰ Tween20的PBS洗，特别是在显色之前要多洗。
12．返蓝：在苏木素复染后，可用碱性缓冲液（如PBS）或Na2HPO4的饱和溶液返蓝。
13．显色：一定要在显微镜下观察，注意控制背景。
DAB法 显示过氧化物酶
〔原理〕 过氧化物酶分解H2O2的过程中，下面反应不直接发生：AH2（供氢体）+H2O2→A（供氢体的氧化物）+2H2O2实验可知，有称为复合物 Ⅰ、Ⅱ、Ⅲ、的酶——底物（受氢体）复合体生成，在复合体zui后分解为酶和水时，游离的原子氧使供氢体氧化而显色。酶组织化学中所使用的供氢体应是含有色原性基团的发色物质，如奈酚和联苯胺。
免疫组化染色步骤（以ABC法为例）
9. Blocking: When goat serum is closed and nonspecific staining is still strong, it can be extended or closed with concentrated serum
10. Antibody dilution: Follow the principle of "Ready-to-use Ready-to-use". Antibody diluted with PBS must be used on the same day.
11. Background High: If the background is still high, under appropriate conditions of antibody concentration, reaction time, reaction temperature, etc., wash with PBS containing 1 ‰ Tween20, especially before developing.
12. Back to blue: In the hematoxylin counterstain, available alkaline buffer (such as PBS) or Na2HPO4 saturated solution back to blue.
13. Color: Be sure to observe under the microscope, pay attention to control the background.
DAB method shows peroxidase
[Principle] In the process of decomposition of H2O2 by peroxidase, the following reaction does not directly occur: AH2 (hydrogen donor) + H2O2 → A (hydrogen donor oxide) + 2H2O2 experiment shows that there are complex Ⅰ, Ⅱ , Ⅲ, the enzyme (substrate) (substrate) is generated by the complex, the final decomposition of the complex enzyme and water, the free atomic oxygen oxidation of hydrogen donor and color. Hydrogen donors used in enzymatic histochemistry should be chromogenic substances containing chromogenic groups, such as naloxol and benzidine.
